Introduction: On May 29, 2021, the U.S. FDA announced the accelerated approval of Sotorasib AMG510 for the treatment of non-small cell lung cancer (NSCLC) patients with KRAS G12C mutations. These patients have received at least prior systemic treatment. This is a global anti-cancer therapy targeting specific KRAS gene mutations.

【Indications】

SOTOLDX is a prescription medicine used to treat adults with non-small cell lung cancer (NSCLC):

• Has spread to other parts of the body or cannot be removed by surgery

• Their tumors have an abnormal KRAS G12C gene

• People who have received at least one treatment for cancer before. Your healthcare provider will do tests to make sure SOTOLDX is right for you. It is not known whether SOTOLDX is safe and effective for children.

【Recommended dose】

• Take SOTOLDX exactly as your healthcare provider tells you to. Do not change your dose or stop taking SOTOLDX unless your healthcare provider tells you to.

•Take SOTOLDX once a day at approximately the same time each day.

•You can take SOTOLDX with or without a meal.

• Swallow SOTOLDX tablets whole. Do not chew, crush, or split the tablets.

• If you are unable to swallow the SOTOLDX tablet whole:

o Place your daily dose of SOTOLDX in a 4-ounce (120-ml) glass of non-bicarbonate sodium water without crushing the tablet. Do not use any other liquids.

o Stir until the tablet breaks down into small pieces (the tablet will not be completely dissolved). The mixture may be light yellow to bright yellow in color.

o Take the mixture of SOOTOLDX and water immediately or within 2 hours of preparation. Do not chew the tablets.

o Use an additional 4 ounces (120 ml) of water to rinse the glass and front to ensure you are wearing the full amount of SOTOLDX.

o If you do not mix immediately for pregnancy, mix again before pregnancy.

• If you take antacids, take SOTOLDX 4 hours before or 10 hours after taking your antacid.

• If you miss a dose of SOTOLDX, take that dose as soon as you remember. If it has been more than 6 hours, do not take that dose. Take your next dose at your regularly scheduled time the next day. Do not take 2 doses at the same time to make up for the missed amount.

• If you vomit after taking one medicine, SOTOLDX, take an extra dose. Take your next medicine on your regularly scheduled day.

【Included】

SOTOLDX may cause serious consequences, including:

• Liver problems. SOTOLDX may cause abnormal results on liver blood tests. Your healthcare provider should do blood tests to check your liver function before you start and while you are taking SOTOLDX.

Tell your healthcare provider the right way if you have any physical signs or symptoms of HIV problems, including:

o Yellowing of your skin or the whites of your eyes (non-stop yellow) o Vomiting or vomiting o Bleeding or bruising o Digging or "brown"

o Chest heaviness o Light-colored stools (bowel movements) o Tiredness or weakness o Pain, soreness, or tenderness on the right side of your stomach area (abdomen)

• Lung or breathing problems. SOTOLDX may cause inflammation of the lungs, which can lead to death.

Tell your healthcare provider right away or get emergency medical help if you develop new or uncomfortable shortness of breath, cough, or fever.

If you have an adverse reaction, your healthcare provider may change your dose, temporarily stop, or permanently stop treatment with SOTOLDX. The most common adverse reactions to SOTOLDX include:

o Manhattan o Heart problems o Interception or bone pain o Cough

o Eating meals o Changes in liver function tests o Fatigue o Changes in certain other blood tests

These are not all the possible consequences of SOTOLDX.

【Survivor】

• Store SOTOLDX at 68°F to 68°F (20°C to 30°C) for a short period of time.

• Bottles are fitted with child-resistant caps.

Keep SOTOLDX and all medicines out of the reach of children.
