Indications
Ixazomib is a proteasome inhibitor that is commonly used in the following situations:

Multiple Myeloma (MM) :
In combination with lenalidomide and dexamethasone, for patients with multiple myeloma who have received at least one prior therapy.
Mechanism of action
Ixazomib is an oral proteasome inhibitor that selectively inhibits proteasome activity, interfering with protein degradation and homeostasis in tumor cells, ultimately leading to cell death.
Dosage

Recommended dose :
- 4 mg orally once a week.
- Ixazomib should be taken on days 1, 8, and 15 of a 21-day treatment cycle.

Combination medication :
- It is usually used in combination with lenalidomide (25 mg/day on days 1 to 21) and dexamethasone (40 mg on days 1, 8, 15, and 22).

Directions :
- Take on an empty stomach (at least 1 hour before or 2 hours after a meal).
- Capsules should be swallowed whole and should not be crushed, chewed or opened.
Dosage adjustments :
- If serious adverse reactions occur (such as hematologic or non-hematologic toxicity), the physician may interrupt treatment or reduce the dose to 3 mg or less.

Side Effect Monitoring :
- Common side effects : diarrhea, constipation, nausea, vomiting, fatigue, peripheral neuropathy, thrombocytopenia, rash, etc.
- Serious side effects : hematological toxicity (such as thrombocytopenia, anemia), peripheral neuropathy, abnormal liver function, gastrointestinal toxicity, etc.

Use in special populations :
- Pregnant and lactating women : Contraindicated. Ixazomib may cause fetal harm, and women of childbearing age are advised to take effective contraceptive measures.
- Children : Safety and effectiveness have not been established.
- Elderly patients : Elderly patients may have lower tolerance to drugs and the dose may need to be adjusted with caution.

Other notes :
- Regularly monitor blood routine, liver function and kidney function.
- If the patient has impaired liver or kidney function, the dose needs to be adjusted accordingly.
Drug interactions
- CYP3A inducers : such as rifampicin and St. John's wort may reduce the efficacy of ixazomib and should be avoided in combination.
- CYP3A inhibitors : such as ketoconazole and erythromycin may increase the blood concentration of ixazomib and should be used with caution.
Contraindications
- Patients who are allergic to ixazomib or its excipients are contraindicated.
Storage conditions
- Store in a dry environment at 20°C to 25°C.
- Avoid high temperature, humidity and direct sunlight. Keep out of reach of children.
Important Tips
- Ixazomib should be used under the guidance of a physician with experience in treating multiple myeloma.
